Abstract: | Two factors (IF-I and IF-II) necessary for the initiation of protein synthesis have been partially purified from a 0.5 M KC1 wash of chicken erythroblast polysomes. IF-I mediates the binding of the initiator tRNA and GTP to a 40 S ribosomal subunit, resulting in the formation of a 44 S initiation intermediate. In the presence of IF-II and a suitable RNA template, the 44 S initiation intermediate combines with a 60 S ribosomal subunit to form a functional 80 S initiation complex. The methionyl moiety of the initiator tRNA in the 80 S initiation complex is able to react with puromycin to form methionylpuromycin. |
