Abstract:
  • 1.1. A method for the determination of glycogen has been developed. The procedure involves digestion of tissue in boiling NaOH, deproteinization of the alkaline digest with HClO4, precipitation with alcohol, washing with alcohol, and color production with anthrone reagent.
  • 2.2. Experiments with glycogen added to liver, skeletal muscle, and heart showed mean recoveries of 92 to 101%.
  • 3.3. The method combines the advantages of the classical alkaline extraction technique and the rapid colorimetric anthrone procedure. The possibility of interference from glycoprotein is removed by the perchloric acid treatment.
