脱血红素细胞色素c自发折叠的进一步验证

摘    要:用胰蛋白酶水解结合聚丙烯酰胺凝胶电泳以及纳秒内源荧光衰减谱分析对鸡心脱血红素细胞色素c在透析复性过程的自发折叠现象作进一步确定。结果显示随着透析复性时间的增加,鸡心脱血红素细胞色素c对胰蛋白酶的水解敏感性逐渐下降,内源荧光寿命值增大,与此对应的马心脱血红素细胞色素c没有发生变化。

Abstract:The spontaneous folding characteristics of chicken heart apocytochrome c was further examined by trypsin hydrolysis followed by SDS-PAGE and analysis of the intrinsic fluorescence decay spectra. The results showed that ,in contrast to horse heart apocytochrome c, hydrolysis accessibility of chicken heart apocytochrome c to trypsin was decreasing while the fluorescence lifetime was increasing with increasing in dialysis time. It was suggested that chicken heart apocytochrome c concomitant with the spontaneous partial folding tends to aggregate and its N-terminal may not involve in the spontaneous. folding.
