绞股蓝组织培养的研究

摘    要:绞股蓝外植体在不同激素浓度的MS培养基上培养,经过正交试验选择了四种最适培养基配方。形态观察和同工酶测定结果表明,在MS基本培养基中添加NAA(1.0~2.0mg/L)可促进愈伤组织和根的生长;添加6-BA(2.0mg/L)也能诱导愈伤组织,并能分化丛生芽;而在6-BA(1.0mg/L)中加入低浓度的NAA(0.1mg/L)和2,4-D(0.1mg/L),对诱导愈伤组织和丛生芽的分化有增效作用。经染色体观察和试管苗大田试种,未发现遗传性变异。

Abstract:Explants of Gynostemma pentaphyllum were cultured on MS media. Four optimum conditions were selected by orthogonal test. Morphological observation and coenzyme determination showed that MS+NAA (1.0-2.0mg/L) promoted the growth of callus and root and that MS+6-BA (2. 0mg/L) induced the callus and differentiation of bush buds on MS basic media; however, the later media added with low concentration of NAA (0. 1mg/L) and 2,4-D (0. 1mg/L) further exhibited to have increasing effect on their callus and differentiation of bush buds. There was not any hereditary variation found through the observation of chromosomes and the tube seedlings in field trial.
