鼠毛及脑线粒体DNA片段缺失与增龄的关系

摘    要:以聚合酶链反应(PCR)技术检测不同年龄Balb/c小鼠脑细胞线粒体DNA片段缺失与增龄的关系.发现老年鼠脑细胞线粒体3867bp片段缺失率为50%;而断奶鼠与青年鼠均无此缺失片段出现;用鼠毛为材料进行无损伤检测亦获类似的结果.有人认为线粒体DNA片段缺失率可作为生物衰老的一种生物学标志

Abstract:Deletions in the mitochondrial DNA(mtDNA)of Balb/c mouse cerebrums were detected by PCR.Analysis was carried out on mice of various ages.No deletion was found in Weaning(4 weeks)mice and young(6-12 months)mice.However,mtDNA deletion(3 867 bp)in cerebral cells was present as a 0 63 kb of PCR product in 50% of old(28-32 months)mouse groups.The results indicate that mtDNA deletions are associated with aging.Using hairs of mice as test samples,the results were just the same as above.Therefore this non damage assay of mtDNA deletion seems useful in aging research.
