毛喉鞘蕊花毛状根培养及二萜化合物的形成

摘    要:发根农杆菌pRi15834菌株感染毛喉鞘蕊花的根、茎、叶及小苗,均诱导出毛状根。通过纸电泳,在毛状根中检测到甘露碱。该毛状根不仅具有典型的形态特征,并能合成二萜化合物,对其中3个主要成分forskolin,1,9-dideoxyforskolin和coleol分别进行了定量测定。MS培养基有利于forskolin和coleol的合成,而B5培养基促进1,9-dideoxyforskolin的合成。

Abstract:Hairy root cultures of Coleus forskohlii Briq. were established from intact plant or explants of roots,stems and leaves by infecting with Agrobacterium rhizogenes strain pRi 15834.Transformation was confirmed by paper electrophoresis with mannopine detection. These cultures displayed the typical characteristics of hairy roots.All examined roots formed labdane diterpenoids.Three mainidentified diterpenoids were forskolin, 1,9-dideoxyforskolin and coleol which were measured quantitatively.MS medium favours forskolin and coleol formation, and B 5 medium enhances 1,9-dideoxyforskolin formation.
