高压液相色谱法分离寡糖的对氨基苯甲酸乙脂衍生物

摘    要:寡糖的对氨基苯甲酸乙酯衍生物,以及它的全乙酰化产物具有紫外吸收基团适合HPLC分离,研究了氨基柱和十八烷基柱对寡糖混合物的分离性能及检测灵敏度。从免疫球蛋白M中分离到的寡糖组分,衍生后用HPLC进一步分离提纯出另一些含量较少的糖链。

Abstract:A series of oligosacchrides were briefly derivatized the reducing termini with suitable UV-absorbing moiety (ABEE) and then all particular derivatives were applied to peracetylation, both with and without peracety]ation could be purified by HPLC. We compared with different results of resolution and sensitivity on an aminopropyl-bonded silicon column and C_(18) column. Using this experimental strategy a few minor components isolated from IgM were also obtained and detected. Improved sensitivity and resolution are especially useful for trace quantities of natural oligosacchrides.
