人脑胶质瘤细胞质膜的制备和鉴定

 本文介绍以手术摘除的人脑胶质瘤为标本,应用蔗糖梯度离心建立的一种较为简便的人脑胶质瘤细胞质膜制备法。经标记酶测定,化学组成分析及电镜检查,证实质膜具有一定的纯度。质膜中5’-核苷酸酶(质膜标记酶)活性为全匀浆的8.2倍,其它亚细胞组份污染较小。质膜收量为24.6％。生化测定还显示质膜5’-核苷酸酶活性随着胶质瘤恶性程度升高而下降。

PREPARATION AND IDENTIFICATION OF PLASMA MEMBRANE FROM HUMAN BRAIN GLIOMA

A procedure was developed by means of sucrose density gradient centrifuga-tion to isolate plasma membrane of human brain glioma. 12 gliomas were studied. The purity of the membrane was checked by the marker enzyme examination,chemical analysis and electron microscope morphological observation.The 5'-nucleotidase and Na K -ATPase (plasma membrane marker) activities were enriched 8.2 and 7.6 folds respectively in purified plasma membrane and the contamination of other subcellular fractions were low.The yield of plasma membrane is about 24.6% of total 5'-nucleotidase activity.5'-nucleotidase activity is lower in malignant glioma than benign glioma. This membrane fraction can be used in clinical and basic investigation.
