| 摘 要： | 我们设计了一种分离小鼠肝细胞的简易灌流法。它具有操作简便,不需特殊装置,胶原酶耗费少等优点。在总活细胞产量及每100g体重活细胞产量等方面显著高于振荡法(P<0.001),而接近Seglen灌流法。此外,对经本法分离的肝细胞及肝非实质细胞进行培养,获得了良好效果。

| Abstract: | A simplified perfusion method in isolating mouse liver cells was introduced. It is easy to be operated and no special instruments are needed. Moreover, collagenase consumption is much less with this method than with routine method Total viable cells isolated and total viable cells per 100 g body weight in the group using simplified perfusion method were significantly higher than in the group with shaking method(P< 0.001), and were similar to those in the group with Seglen's perfusion method. In addition, the hepatocytes and nonparenchymal liver cells isolated with the simplified perfusion method grew quite well during incubation. |
