具有中等毒性的马氏钳蝎神经毒素的纯化和初步晶体学研究

运用柱层析技术对产自淅川和常德的马氏钳蝎毒素进行分离纯化，得到８种哺乳动物神经毒素。运用制备型等电聚焦电泳技术，对常德样品中具有中等毒性的蝎神经毒素（ＢｍＫ５）进一步纯化，获得了高纯度样品。两个产地的蝎毒素ＢｍＫ５均已经成功地获得了大晶体。空间群均为Ｐ２１２１２１，晶胞参数分别为：ａ＝３８．４６埃，ｂ＝３７．２８埃，ｃ＝３６．９７埃（淅川）；ａ＝３８．４４埃，ｂ＝３７．５５埃，ｃ＝３６．８３埃（常德）。对两个产地的晶体分别收集了２．１埃（淅川）和１．６２埃（常德）分辨率的衍射数据。

Purification, Crystallization and Preliminary Crystallographic Analysis of a Neurotoxin with Medium Toxicity from Chinese Scorpion(Buthus martensii Karsch)

BmK5,a neurotoxin with medium toxicity to mice from the venom of Chinese scorpion (Buthus martensii Karsch) in Xichuan and Changde of middle China,has been purified and crystallized and crystals good enough for X-ray analysis at high resolution have been obtained. The space group is P212121.The cell parameters are a=38.46A,b=37.28A,c=36.97A (Xichuan crystal) and a=38.44A,b=37.55A,c=36 .83A(Changde crystal);the unit cell volume is about 53 kA3.The X-ray diffraction data of Xichuan crystal have been collected at 2.1A resolution and the data at 1.62A resolution of Changde crystal have also been collected.BmK5 and two other neurotoxins,BmK1 and BmK5 which possess strongest and weakest toxicity in BmK series respectively and have been crystallized,also form a series of scorpion neurotoxin crystals with different texiejties.Evidently,the X-ray analysis and the comparison of their structures will provide significant information for understanding the structure-function relationship of the scorpion neurotoxins.
