摘    要:RT-PCR、锚定PCR、cDNA末端快速扩增(RACE)、step out RACE、DDRT-PCR、cDNA代表性差异分析、抑制性削减杂交等都是建立在PCR基础之上的cDNA分子克隆技术。本介绍各种技术的优缺点、应用以及与同类技术进行比较,这些技术现在已经成功应用于大量EST的分离、cDNA库的构建、编码产物未知的差别表达基因的分离以及全长cDNA基因序列的克隆等方面。

Abstract:RT-PCR, single-sided(anchored) PCR, RACE, step out RACE, DDRT-PCR, cDNA RDA and SSH are all the techniques of cDNA cloning based on PCR. The advantages and disadvantages of each technique were dicussed and compared with those of their counterparts. These techniques are now successfully used in the fields of isolation of numerous ESTs, construction of cDNA libraries, obtaining of differentially expressed new genes and cloning of cDNA in full length.
