摘    要:以克隆的人巨细胞病毒(Human Cytomegalovirus,简称HCMV)DNA片段做探针,用核酸杂交方法检查了50例北京地区幼儿园正常儿童的尿标本。查出HCMV DMA阳性者24例,占48%。在两个群体和两个年龄组之间,阳性检出率有显著性差异。利用探针可查出同源DNA5~10pg,而不与其它病毒或细胞DNA杂交。标记不同的探针,使用不同的固定方法,其敏感性不同。此法可在分子水平上查出HCMV基因,比分离病毒更迅速敏感。

Abstract:50 urine specimens from the kintergarden children in Beijing were exa-mined by using DNA hybridizatioa test with cloned HCMV probe. 24 out of 50 samples shown positive result. The positive rate is 48 per cent.Different positive rates between the 2 kiatergardens and the 2 age groups were observed. The probe can detect homologous DNA at pg Ievel and specifically hybridized with DNA of HCMV but not with other virai or cell DNAs tested.The DNA-DNA hybridization test is more rapid and more sensitive thaa the virus isolation method. The assay will be useful for rapid virai diagn osis and clinical research.
