短梗霉细胞内黑色素的分离提取

摘    要:采用单因素试验和正交试验对短梗霉细胞内的黑色素提取条件进行了优化,结果表明:在pH为2~3、温度为70℃、NaOH浓度为1.5mol/L、发酵液与浸提液之比1:1(V)的条件下黑色素的提取量可达2.2g/L。对提取的黑色素进行了纯化,研究了其紫外和红外光谱学性质,紫外光谱图显示随波长的减少其吸收值增大,在215nm处有特征吸收峰;红外光谱图在3和6μm处有吸收峰,证实短梗霉黑色提取物是一种以芳香环为结构主体的异聚体黑色素。

Abstract:Single factor and orthogonal experiments were used to optimize the extraction conditions for melanin from Au-reobasidium pullulans.The result showed that on the optimum conditions of pH 2-3 ,temperature 70℃ ,sodium hydroxide 1 mol/L,the ratio of fermentation liquid to alkaline solution 1:1 (V) ,each liter of fermentation liquid yielded 2.2 g of melanin.After extracted and purified, the UV absorption spectrum of the melanin showed a steadily increasing absorption ranging from 300 nm to 200 nm with a peak at about 215 nm.In the infra-red spectrum of the melanin,there were char-acteristic absorption at about 3 and 6 μm.The above results verified that the product we had extracted was indeed a kind of melanin which is a polymer of aromatic heterocylic structure.
