摘    要:构建叶绿体超氧化物歧化酶基因(ChlSOD),采用RT-PCR方法分离豌豆RUBP羧化酶小亚基导肽基因(TP),定向克隆至pUC19测序,定向克隆烟草MnSOD成熟蛋白基因(SODm)至pUC19;采用平粘端连接法将二者在pUC19中构成嵌合基因ChlSOD,并对此基因进行序列分析,序列分析表明:TPcDNATP,12bp的Linker及615bp SODm。TP与ChlSOD基因的序列分析与国外报道序列完全吻合。

Abstract:To construct the chimeric gene of chloroplast manganese superoxide dismutase(ChlSOD).The gene coding ribulose-1.5-bisphosphate carboxylase/oxygenase small subunit precursor(TP) from pea has been isolated by PT-PCR,inserted into pUC19 and sequenced; the gene encoding MnSOD mature protein (SODm)from Nicotana. tubaccum has been cloned into pUC19; a chimeric DNA fragment consisting of these two genes is constructed into pUC19 by sticky-blunt end ligation and sequenced. The cDNA sequence of TP is 171 bp,encodes 57 amino acids; the open reading frame of ChlSOD is 798 bp, encodes 266 amino acids, which includes 171 bp TP,12 bp linker, 615 bp MnSOD mature protein gene. The sequence analysis of these two genes is completely identical with those have been reported.
