转抗盐基因八里庄杨组培苗快繁研究

摘    要:以转1-磷酸甘露醇糖醇脱氢酶基因的八里庄杨为试材,筛选出了快繁的一系列培养基。分化培养基MS+BA0.3mg.l^-1 IBA0.1mg.l^-1分化苗多,苗头密度高,叶色绿,并且可以节约对KT的使用。伸长培养基KT2.0mg.l^-1+NAA0.1mg.l^-1使苗子伸长现象明显,苗子长势壮,叶子肥大。生根培养基以MS+IBA0.2mg.l^-1为最佳,8天后开始生根,数量较多,生根后的试管苗生长粗壮,有利于移栽成活。以简化生根技术对杨树转化体进行生根试验,瓶外生根苗的移栽成活率明显比瓶内高,组培苗已成功移栽于大田中。

Abstract:Poplar transferred with mtl-D gene was used as the experimental material . A series of mediums for micropropagation were obtained. The optimum buds differentiation was the medium with MS+BA0.3mg.l-1+IBA mg.l-1, which induced greener leaf, more buds and higher bud density.;The medium with MS+KT2.0 mg.l-1+ NAA mg.l-1 was best for bud elongating, in which bud elongated obviously and had higher growth potential and bigger leaves;The medium with MS+IBA0.2 mg.l-1 was best for seedling rooting, which induced seedlings to be rooted after 8 days with lots of roots. After being rooted, the strongest seedling was best for transplanting. Transgenic poplar was also rooted with simplified rooting method with higher transplanting survival ratio than that in bottle.
