Abstract:A preparation and stain method of thin sections for small samples with hard, thick shells and soft internal tissues is dealt with in this paper. The results showed that the seeds of Idesia Polycarpa var. vestita Diels at different stages of development, after being immersed in concentrated sulfuric acid for different time to increase the permeability of the seed coats, can be easily prepared into thin sections with whole seed structure by usual method, and that staining in Methylene Blue and Eosin, various cell components can be respectively stained different colors.
