红斑性肢痛症相关痘病毒血清免疫学特征的研究

摘    要:本文从四个方面综合报道了红斑性肢痛症相关痘病毒(ERPV)血清免疫学特征的研究结果。血清学鉴定表明,ERPV为正痘病毒属内成员,但与正痘病毒属内成员痘苗病毒和鼠痘病毒具有中和抗原表位差异。流行性红斑性肢痛症患者血清中含有抗ERPV的中和抗体和抗ERPVA型包涵体IgG抗体,其检出率明显高于当地未发病者和美国人群的检出率。

Abstract:The serological reactivity of erythromelalgia-related poxvirus (ERPV)which originally was isolated from the throab swab of a patient with eryhth-romelalgia was analyzed in the present study. Rabbits immunized by foot-padinoculation of ERPV started to produce neutralizing (NT) antibody to ERPVat 2 weeks postinfection (p. i.). The peak NT antibody titers were observedat 4 weeks p. i. and then began to decline. When eight antisera including sheepanti-Orf, bovine anti-parapox, rabbit antivaccinia, rhesus monkey anti-Tanapox,rabbit anti-Yabapox, raccoon anti-raccoonpox, human anti-molluscum contagio-sum, and mouse antiectromelia virus were used to identify 6 ERPV isolates,only rabbit anti-vaccinia, mouse anti-ectromelia, and rabbit anti-ERPV anti-sera reacted with all isolate. Thus, ERPV was serologically identified as anorthopoxvirus. However, when rabbit anti-ERPV antisera were useed to cross-neutralize vaccinia or ectromelia virus, neither was neutralized by these anti-sera, suggesting that there are some epitope differences in NT antigens bet-ween ERPV, vaccinia and ectromelia. Furthermore, 68% (17/25) of thepatients with a history of epidemic erythromelalgia and only 25% (2/8) of non-disesed control subjects tested positive for anti-ERPV antibody in their serausing a plaque reduction NT test. In addition, the detection of anti-ERPVIgG using immunofluorescence was significant higher (P<0.005) in the serafrom patients with epidemic erythromelalgia (49/125, 39.2%) as comparedwith the sera controls (2/17, 11.8%)or "American" controls (5/42, 11.9%).
