Abstract:A protocol for the in vitro germination and the establishment of tissue cultures of Bulbine caulescens Linn., Kniphofia pumila Kunth, and K. uvaria Hook has been developed. The members of the family Asphodelaceae contain interesting substances with antiplasmodial activity against several strains of Plasmodium falciparum. With respect to species of Bulbine, this is the first time that in vitro germination has been achieved. For seeds of K. pumila and K. uvaria, a facilitated germination was attained by an optimized pretreatment of the seeds. After in vitro germination, root explants of Kniphofia and bulb explants of B. caulescens were successfully transformed into callus cultures. From these, liquid cultures of the two species of Kniphofia were easily produced. Studies on shoot multiplication of Kniphofia in vitro were also performed.
