摘    要:目的探讨苏丹红对小白鼠血液和组织器官的影响。方法取小鼠连续12 d腹腔注射不同剂量苏丹红Ⅰ、Ⅱ、Ⅲ、Ⅳ(0、40、80、160 mg/kg),末次给药24 h后采集血样,用血细胞分析仪检测血常规指标,毛细血管法测定凝血时间,剖腹取主要器官称重,计算脏器系数。结果苏丹红Ⅰ和Ⅱ组血液红细胞数明显减少,而苏丹红Ⅲ和Ⅳ组不明显,苏丹红不同剂量之间比较,低剂量苏丹红Ⅰ和高剂量苏丹红Ⅱ组比苏丹红Ⅲ和Ⅳ组血液红细胞数变化明显;苏丹红Ⅰ和Ⅱ组血液血红蛋白含量明显降低,而苏丹红Ⅲ和Ⅳ组血红蛋白含量变化不明显。四种不同剂量苏丹红组小鼠血液中血小板数量减少,随着剂量增加小鼠血液凝固时间延长。随着苏丹红剂量增加,苏丹红I和II组小鼠血液白细胞数比对照组明显增加,苏丹红Ⅲ和Ⅳ组白细胞数有增加趋势但差异不显著。与对照组比较,苏丹红组小鼠血液白细胞数明显增加、而淋巴细胞和单核细胞显著减少。随着苏丹红注射剂量增加,肾脏和脾脏脏器系数较对照组明显增大,肝脏脏器系数变化不明显。结论苏丹红对小鼠血液细胞、血红蛋白、血小板和凝血时间以及组织器官都有不同程度影响。

Abstract:Objective The aim of this study was to investigate the in vivo effects of Sudan red dyes on the blood and organs in mice. Methods The mice were intraperitoneally injected Sudan red Ⅰ , Ⅱ , Ⅲ and Ⅳ in a dose of 0, 40,80, or 160 mg/kg for 12 consecutive days, respectively, 12 mice in each group, male:female 1: 1. Blood samples were collected at 24 hours after the last administration, and blood routine tests were conducted using a blood cell analyzer. The blood clotting time was determined by capillary method and organ coefficients were calculated by weighing separated organs from the mice. Results The counts of erythrocytes in the Sudan red Ⅰ and Ⅱ groups were significantly reduced but not significantly changed in the Sudan red Ⅲ and Ⅳ groups. Compared between different doses of Sudan red, there were significant changes in the counts of blood red ceils in the low dose Sudan red Ⅰ group and high dose Sudan red Ⅱ group than that in the Sudan red Ⅲ and Ⅳ groups. The blood hemoglobin contents in the Sudan red Ⅰ and Ⅱ groups were significantly reduced but were not changed in the Sudan red Ⅲ and Ⅳ groups. There was a reduced count of platelets in the four Sudan red groups of different doses, and blood coagulation time was extended with the increasing dose of Sudan red. The leukocyte counts were significantly increased in the Sudan red Ⅰ and Ⅱ groups compared with that of the control group, along with the increase of Sudan red doses. There was an increasing trend of white blood cell counts in the Sudan red Ⅲ and Ⅳ groups, but the difference between two groups was not significant. Compared with the control group, blood leukocyte count was significantly increased, and the counts of lymphocytes and monocytes significantly reduced in the Sudan red groups. With increasing dose of Sudan red, the organ coefficients of the kidney and spleen were significantly increased compared with that of the control group, but the organ coefficient of the liver was not significantly changed. Conclusions The results show that Sudan red dyes have an impact to a varying degree on mouse blood cells, hemoglobin, platelets, clotting time, and some visceral organs.
