拟南芥叶绿体分裂突变体cpd111的基因定位与分析

摘    要:通过EMS(ethyl methane sulphonate)诱变从拟南芥(Arabidopsis thaliana)突变体库中筛选到一个叶绿体分裂突变体(c)hloro(p)last (d)ivision 111 (cpd111).遗传学分析表明,该突变体的表型是单基因控制的隐性性状.与野生型相比,突变体植物细胞的叶绿体数量少,叶绿体形态和大小多样化,并且细胞体积与叶绿体数量之间无相关性.利用图位克隆的方法确定cpd111的突变基因为FtsZ1.进一步的分析表明,该突变影响FtsZ7基因mRNA的正常剪切和稳定性,使蛋白质翻译提前终止,最终导致叶绿体分裂异常.该工作为研究FtsZ1在叶绿体分裂中的作用提供了新的材料和线索.

Abstract:Ethyl-methane sulfonate mutagenesis revealed a chloroplast division mutant, chloroplast division111 (cpd111), in Arabidopsis thaliana. Genetic analysis indicated that the mutant was controlled by a single recessive gene. Chloroplasts in the mutant were fewer in number and more heterogeneous in size than those in the wild type, The corre- lation between chloroplast number and cell size was not obvious in the mutant. Map-based cloning revealed a mutation in FtsZ1 responsible for the mutant phenotype. The mutation affected the splicing and stability of FtsZl mRNA, caused early translational termination of the protein and resulted in a chloroplast division defect in cpd111. We provide new materials and information for a better understanding of the function of FtsZ1 in chloroplast division.
