GM_1激活环核苷酸磷酸二酯酶的作用

摘    要:神经节苷脂GM1能激活CaM依赖性环核苷酸磷酸二酯酶,其AC50为1.56μg/ml.这种作用并不一定依赖Ca2+的存在.在有Ca2+存在时,GM1对PDE的最大激活活性低于CaM,仅为CaM的80.4%;没有Ca2+存在时,GM1与CaM对PDE有同样的最大激活活性(100%).GM1能使CaM对PDE的激活曲线左移,AC50降低,但不改变CaM对PDE的最大激活活性.三氟啦嗪能使GM1激活的PDE的活性降低,其IC50为16.3μmol/L.

Abstract:Ganglioside GM1 can activate calmodulin dependent cyclic nucleotide phosphodiesterasein the presence or absence of Ca2+. The AC50 of GM1 on PDE is 1. 56 μg/ml. The highest activityof PDE stimulated by GM1 is the same as CaM and is 80. 4 % of PDE activated by CaM in the absence of Ca2+. In the presence of GM1, the activity curve of CaM on PDE moved to the left, theAC50 of CaM on PDE is decreased and the concentrations of CaM at the highest activity of PDE isalso decreased. TFP can antagonize the GM1 to stimulate PDE. The IC50 of TFP on GM1 is 16. 3μmol/ L.
