| Abstract: | Hepatitis B surface antigen S and preS1(21-47) coding sequences were amplified by PCR and the preS1 sequence was fused to the 3' end of S region. The chimeric construct was sequenced and cloned into Pichia pastoris expression vector pAO815. The expression plasmid was transformed into Pichia strain GS115 by electroporation. A high-level expression strain(GS115-SS1) was established by primary screening for His~ + transformants and further analysed for induction products. ELISA results demonstrated that the expressed protein had S and preS1 antigenicity simultaneously. The titer of RPHA reached 1∶2048, corresponding to 32 g/ml. The molecular weight of the fusion protein was 28kD as verified by Western blot and was identical to the theoretical size. |
