Abstract:To establish an HPLC method for determination of chlorogenic acid in cucumber skin,Zorbax Eclipse XDB-C18 was adopted.The mobile phases consisted of CH3OH-0.04% H3PO4(25∶75) at a flow rate of 1.0 mL·min-1,the detection wavelength was set at 326nm,and column temperature was 30 ℃.The linear range of chlorogenic acid was 0.0111~0.1332mg·mL-1(r= 0.9992).The average recoveries(n=5) was 99.97% and RSD was 2.07%.The contents of chlorogenic acid in cucumber skin were more than 8.10 mg·g-1.The method was simple,rapid and accurate with a good reproducibility,and could be applied to determine of chlorogenic acid in cucumber skin.
