摘    要:应用500~2500伦琴(R)的60钴γ射线照射滇紫草(OnosmapaniculatumBur.etFranch.)细胞系,确定了应用γ射线处理促进紫草素合成的最适辐照剂量为1500R,处理后的细胞系在生产培养基上培养21d后紫草素含量(以干重计)达到94.79mg/g,较对照提高了1446%,通过小团块选种法从中筛选到了色素含量(以干重计)高达103.42mg/g的高产细胞系Mut1,其营养生长与对照相比没有差别。

Abstract:The effect of different dosages 500 R, 1 000 R, 1 500 R, 2 000 R and 2 500 R of gamma ray on the shikonin formation in cultured cells of Onosma paniculatum Bur. et Franch. is reported. It is determined that the optimum dosage to stimulate the formation of shikonin is 1 500 R, a 144.6% increase of shikonin content was observed in the irradiated callus, with the 94.79 mg/g (dry weight) of shikonin content. Furthermore, a high production cell line Mut 1 with 103.42 mg/g (dry weight) of shikonin content was selected from the callus, there was no significant difference in growth between Mut 1 and the control.
