Abstract: | Two major approaches of genetic transformation mediated by pollen are to use microspore embryogenesis as recipient system and take pollen as mediator of exogenous DNA to produce transgenic seeds after artificial pollination. A successful procedure of pollen mediated transformation in tobacco includes following process bombarding mature pollens with microprojectile bombardment or germinating pollens under electroporation, followed by fertilizion with the exogenous DNA containing pollens to produce transgenic seeds. Therefore, in order to get high frequency of stable transformant, the present work attempted to reduce nuclease activity during pollen germinating, select pollen cell growth state for exogenous gene to enter genital cell, set up a highly efficient microfertilization technique and select transformant ahead. |
