用线粒体DNA鉴定玉米雄性不育细胞质的研究

摘    要:参照Kemble法,通过分析玉米线粒体DNA,鉴定出我国生产上应用的玉米双型和唐徐型雄性不育细胞质属于S组。近年新获得的不育类型Y型属于T组,为玉米种子生产合理应用雄性不育细胞质提供了可靠依据,并对鉴别玉米雄性不育细胞质的方法进行了讨论。

Abstract:The modified Kemble's methods were used for the identification of male sterile cytoplasms in maize.The Xho 1 restriction patterns of mitochondrial DNA from Shuang A and Tang A cytoplasms,which are used in seed production in China,were indistinguished from that of S cytoplasm control.Undigested mitochondrial DNAs from Shuang A and Tang A cytoplasms also clearly showed the S1 and S2 plasmid-like DNAs characteristic of the S cytoplasm group.These data indicated that Shuang A and Tang A cytoplasms belonged to S cytoplasm group.The comparison of restriction patterns of mitochondrial DNAs from Y cytoplasm; which was discovered several years ago,with that T cytoplasm indicated that Y cytoplasm belonged to T cytoplasm group.These results will be helpful to the reasonable arrangement of male sterile cytoplasm in seed production in China.The methods of identification of male sterile cytoplasm in maize were discussed.
