地黄扩展蛋白基因RgExpA1的克隆与表达分析

摘    要:采用RT-PCR方法对9个地黄扩展蛋白基因在地黄不同组织器官、不同发育时期的表达特点进行了分析,在此基础上用RACE方法克隆了主要在根中表达的RgExpA1基因的全长,并对该基因编码氨基酸的功能位点、拼接方式以及与同源基因的进化关系作了初步分析。

Abstract:The expression of nine expansin genes in different organs of Rehmannia glutinosa and different developmental stages of Rehmanniaroot was analyzed by RT-PCR.The RT-PCR result showed that the expression of RgExpA1 was root-specific and RgExpA1 expressed strongly in developing tuberous root.It implied that RgExpA1 was involved in tuberous root development.Based on the cDNA fragment sequence,the full length cDNA of RgExpA1 was cloned using RACE(rapid amplification of cDNA ends)technology.We also studied the splicing sites of RgExpA1 and the active sites of its product.The homologous relationship of RgExpA1 with other α-expansins and its putative function were also analyzed in this research.
