金银花黄酮醇合成酶基因全长克隆及其序列分析

摘    要:根据已知的其它物种黄酮醇合成酶cDNA保守序列设计引物,用RT-PCR技术从金银花叶片中扩增获得黄酮醇合成酶基因部分eDNA序列,再用RACE技术获得其两端序列.序列拼接得到完整的1 248bp的黄酮醇合成酶基因,根据获得的序列,设计引物扩增获得1 001 bp的开放阅读框,编码333个氨基酸.序列分析显示,金银花黄酮醇合成酶与烟草、马铃薯和桔梗的同源性分别为86%、83%和80%,与其它物种的同源性也在80%左右,表明不同物种中黄酮醇合成酶基因具有高度同源性.

Abstract:A pair of primers were designed according to the conserved cDNA sequence of flavonol synthase(FLS)gene of other species and partial cDNA sequence of FLS was cloned fromFonicera japonicaleaves using RT-PCR and RACE.The results indicated that the full length cDNA sequence of FLS was 1 248 bp and it included an open reading frame(ORF)which encoded 333 amino acids.Sequence analysis showed that the homologies of FLS cDNA from Fonicera japonica withNicotiana tabacum,Solanum tuberosum andPlatycodon grandiflorus were 86%,83%,80% and among the others were about 80%,which revealed that the FLS gene kept highly conserved among different species.
