干细胞相关因子在子宫内膜息肉中的表达

摘    要:探讨ipo13、c-kit、CD146、bcl-2和bax在子宫内膜息肉(endometrial polyp,EP)和正常内膜组织中的表达差异及临床意义。收集本院2010年3-7月行宫腔镜手术取得的40例子宫内膜息肉(病例组)与40例正常内膜组织(对照组)。采用实时荧光定量PCR技术(RT-PCR)检测ipo13、c-kit、bcl-2和bax mRNA的表达;免疫组织化学S-P法检测ipo13、CD146、bcl-2和bax蛋白的表达。无论在月经周期的增生期或分泌期,EP中ipo13、c-kit、CD146和bax mRNA和蛋白的表达均低于同期正常子宫内膜组织,差异有统计学意义(P<0.05),bcl-2均比同期正常子宫内膜增加,差异有统计学意义(P<0.05)。子宫内膜干/祖细胞活性异常和子宫内膜凋亡减少与子宫内膜息肉发病有关,其中子宫内膜干/祖细胞活性异常使内膜凋亡减少可能是EP发病的主要因素。

Abstract:It was to investigate the expression of ipo13,c-kit,CD146,bcl-2 and bax in endometrial polyps and normal endometrial tissues and its clinicopathologic significance.40 specimens of endometrial polyps tissues(case group) and 40 normal endometrial tissues(control group)in our hospital from March to September in 2010 were collected.Expression of ipo13,c-kit,CD146,bcl-2,bax mRNA and protein was detected by real-time quantitative polymerase chain reaction(RT-PCR) and immunohistochemical staining in all specimens.Whether in the proliferative phase or secretory phase of the menstrual cycle,the expression of ipo13,c-kit,CD146 and bax mRNA and protein in EP was significantly lower than normal endometrial tissues(P<0.05) and the expression of bcl-2 mRNA and protein expression was higher in EP than normal endometrial tissues in the same period(P<0.05).The expression of ipo13,c-kit,CD146,and bax mRNA and protein in endometrial polyps is significantly lower than that in normal endometrial tissues.While,bcl-2 expression up-regulated in polyps.Our results suggested that endometrial stem/progenitor cells dysfunction and endometrial cells apoptosis in decreasing related to endometrial polyps,endometrial stem/progenitor cells dysfunction causing endometrium apoptosis reduction may be the main reason for the occurrence and development of endometrial polyps.
