应用RAPD技术鉴别微生态菌种

摘    要:目的:建立应用RAPD技术鉴别微生态菌种的方法,提高菌种鉴别水平。方法:应用RAPD(随机扩增多态性)方法,选用5条随机引物,利用PCR方法,对3株长双歧杆菌,3株嗜酸乳杆菌,3株粪肠球菌进行基因组DNA指纹图谱分析。结果:发现不同菌株扩增的DNA片段的大小、数量是有差异的。结论:此方法具有可重复性,方便、快速和准确的优势,可用于微生态制剂生产用菌株的鉴别。

Abstract:Objective:Physical bacteria in the microbiological products always have the similar phenotype,their characterization is difficult by using the classical methods.Methods:RAPD conditions were optimized to generate reproducible banding patterns by testing primers,thermocyclers and overall reproducibility it repeat DNA analysis and separate DNA extractions.Five primers were chosen and distinguished among strains of 3 strains of Bifidobacterium ,3 strains of Lactobacillus acidophilus, 3 strains of Enterococcus faecium. Results:This method was highly reproducible in repeated analysis.Differences between the strains were evaluated based on their RAPD profiles.Furthermore,application of the RAPD technique may also be useful and faster,than traditional systmatics for placement of industrial strains into specific clusters.Conclusion:It is very useful in differentiating the difference of the strains,identifying the specialty and originality of the strains.
