Abstract:The addition of mercaptoethanol to the culture medium has permitted a mixed-lymphocyte culture (MLC) reaction to be achieved with mouse spleen cells in a chemically defined protein-free medium. Enhancement of the MLC by unrelated reducing agents indicates that the active moiety of MET is the sulfhydryl group. Although the mode of action of mercapto-ethanol is yet to be elucidated, it appears that the time at which it affects the reaction is during the first 24 hr of culture.
