bFGF对血管性痴呆大鼠海马神经干细胞的影响

摘    要:目的皮下注射bFGF于血管性痴呆大鼠,研究用药前后对大鼠海马神经干细胞增殖能力的影响。方法制作VD大鼠模型,随机取用VD大鼠模型12只,分治疗组6只,痴呆组6只。另外,取假手术组6只。皮下注射bFGF于治疗组中血管性痴呆大鼠。治疗5周后,以Morris水迷宫定位航行试验和空间探索试验来检测大鼠的学习记忆能力,巢蛋白(nestin)免疫组织化学染色,观察海马nestin阳性细胞数的变化。结果治疗组大鼠海马nestin阳性细胞数较痴呆组明显增多。结论皮下注射bFGF后能迁移至海马,诱导海马产生nestin阳性细胞,刺激大鼠海马神经干细胞增殖,修复受损组织。

Abstract:Objective To study the change of the neural stem cells in hippocampus of vascular dementia rats after hypodermic injection of bFGF. Methods After the vascular dementia model was reproduced, the 12 model rats were randomly divided into therapy, dementia group. 6 rats were taken in pseudo-operative group. The vascular dementia rats in therapy group were treated by hypodermic injection of bFGF. After 5 weeks, abilities of learning and memory of three section rats were tested by using the Morris water maze. Immunochemistry staining was used to observe the number of nestin positive neurons in hippocampus. Result The number of the nestin positive neurons in the CA1 subfield of the hippocampus were more significantly increased in therapy group than that in dementia group. Conclusion bFGF by hypodermic injection can migrate to hippocampus area and induce hippocampus to create nestin positive neurons, bFGF can stimulate hippocampus to generate neural stem cells and repair damaged tissue.
