NGI重组DNA疫苗对转基因AD小鼠的治疗作用

摘    要:目的研究NGI对转基因阿尔茨海默病小鼠的治疗作用。方法构建包含小鼠OMgp细胞外8LRP重复序列,人Tenascin-R的EGF-L结构域,以及人Nogo-A的氨基端和66个氨基酸细胞外域的重组质粒,肌肉注射AD小鼠,空载体组和空白对照组为对照组。水迷宫检测小鼠行为学差别,免疫组化检测其病理差别。结果4.5月龄AD小鼠经过水迷宫隐蔽平台实验,2d后疫苗组小鼠潜伏期比对照组缩短30%(P〈0.05)。探索实验显示3月龄疫苗组比空载体组和空白对照组小鼠穿越目标象限的次数分别增加48%和44%(P〈0.05)。结论NGI重组DNA疫苗能够提高转基因AD小鼠的学习与记忆能力。

Abstract:Objective To study the therapeutic effects of NGI on transgenic AD mice. Methods The recombinant plasmid construct includes cDNA sequence of the extracellualar 8LRP repeats of mouse OMgp, EGF-L domain of human Tenascin- R,and N-terminal and 66-amino acid extraceUular domains of human Nogo-A. The AD mice were intramuscularly injected with plasmid., empty vector and blank group as control group. The behavioral tests were examined by the water maze tests. The pathologic difference was observed by immunohistochemistry. Results The average latency to find the hidden platform after 2 days in the water maze tests of vaccine group of 4.5-month APP/PS1 double transgenic mice is 30% shorter than the control group. The Annulus crossing index of the 3-month mice of the vaccine group in probing test is 48% and 44% more than the empty vector and blank group. Conclusion NGI recombinant DNA vaccine can improve the leaming and memory ability of transgenic AD mice.
