小鼠生态条件对Friend小鼠白血病病毒感染的影响

摘    要:本文探讨了F1小鼠生态条件(鼠龄和免疫状态)对Friend小鼠白血病病毒(Fr.MuLV)感染的影响。利用对Fr.MuLV抗性的Fv4小鼠(含Fv4抗性基因纯合子的BALB/C小鼠)与对Fr.MuLV敏感的BALB/C小鼠交配,获得含Fv4抗性基因杂合子的F1小鼠(BALB/C×Fv4)。然后经小鼠腹腔接种Fr.MuLV病毒液进行攻击,并检查不同鼠龄和免疫状态下的F1小鼠对Fr.MuLV的敏感性,以阐明F1小鼠的鼠龄和免疫状态对Fr.MuLV感染的影响。结果表明,新生F1小鼠可发生Fr.MuLV感染,脾脏中有大量病毒增殖,并引起白血病;免疫抑制剂Fk506投与下的成年F1小鼠亦可发生Fr.MuLV的感染,脾脏中有病毒增殖,但不引起白血病;而新生或成年Fv4小鼠以及成年F1小鼠可抵抗Fr.MuLV的感染,脾脏中无病毒增殖,不发病。说明F1小鼠的鼠龄和免疫状态等生态条件可影响其对Fr.MuLV感染的敏感性,并且可影响Fv4基因抗病毒作用。

Abstract:The effect of F1 mouse ecological elements that include age and immune status on the infection of Friend Murine leukemia virus(Fr.MuLV) had been investigated in the paper.F1 mice (Fv 4 antiviral gene heterozygote) were the offspring of Fr.MuLV resistant Fv 4 mice (Fv 4 antiviral gene homozygote) and Fr.MuLV sensitive BALB/C mice.Fr.MuLV was injected intra abdominally to the newborn F1 mice and adult F1 mice which had been given immune inhibitor Fk506.Result showed that Fr.MuLV could infect newborn F1 mice,and Fr.MuLV could propagated in mice spleen,and caused leukemia;Fr.MuLV could also infect adult F1 mice which had treated with immune inhibitor Fk506.Whereas the newborn or adult Fv 4 mice,and adult F1 mice could resistant the infection of Fr.MuLV,no viral propagation could be observed in mice spleen.This result suggested the sensitivity of F1 mice on infection of Fr.MuLV was determined by murine ecological elements such as age and immune status.This antiviral effect of Fv 4 antiviral gene was influenced by age and immune status of mouse.This result provides experimental evidence for clarification of the antiviral mechanism of organism.
