抗人铁蛋白单克隆抗体的制备及其在肝癌病人血清铁蛋白测定中的应用

铁蛋白是一种肿瘤相关蛋白质。我们从人肝癌组织中分离纯化了铁蛋白，并筛选到一株抗该铁蛋白的单克隆抗体杂交瘤细胞株（６Ｄ６），它针对铁蛋白上的构象决定簇，并对人源铁蛋白高度专一。然后我们用此单抗建立了测定铁蛋白浓度的夹心ＥＬＩＳＡ法，并对方法的灵敏度，就确度及特异性作了研究。本法用于测定不同血清标本的结果表明：肝癌病人血清中的铁蛋白浓度明显高于正常人，这可能对临床诊断会有使用价值。

Preparation of Anti-ferritin Monoclonal Antibody and Its Application to Measuring Ferritin Concentrations in Liver Cancer Sera

Ferritin is one tumor-associated protein. We purified ferritin from human liver cancer tissues and obtained one antiferritin monoclonal antibody (Mo Ah)6D6. This MoAb reacted with conformational epitopes on ferritin molecules and was highly speeific for human ferritin. Then we developed a sandwich ELISA,where the first antibody was 6D6 and the second antibody was 6D6-HRP, to measure ferritin eoneentrations in sera from liver cancer patients as well as from normal people. The results showed that concentrations of serum ferritin from patients were markedly higher than those from normal people, which may be helpful to clinical diagnosis of liver cancer. In the meantime, the sensitivity and precision of this method were analysed.
