枸杞组织培养中过氧化物酶和可溶性蛋白质的变化

摘    要:枸杞(Lycium barbarum L.)叶组织切块接种在附加6-BA 0.5毫克/升和NAA 0.5毫克/升的MS固体培养基上可大量诱导产生愈伤组??织,培养至30天分化出芽,到50天长成小苗。过氧化物酶活性和可溶性蛋白质的含量分别在大量分生细胞团形成和芽形成时出现两个峰值,酸性蛋白质的种类也在这两个时期迅速增加。过氧化物酶同工酶阳极端出现的四条酶带中,A_2、A_3较稳定,A_1、A_4则在不同发育时期呈现规律性的变化。

Abstract:Leaf explants of Lycium barbarum L,cul-tured on MS medium with 0.5 mg/l BA and0.5 mg/l NAA produced calluses.The budswere differentiated after 30 days,and the re-generated plants were formed after 50 days.Cytohistological observation indicated that cal-lus mainly originated from mesophyll cellsand epidermis cells.In the course of forma-tion of the callus,starch was accumulated innon-divided mesophyll cells and some paren-chyma cells.The clusters of thacheary ele-ments developed in the callus before formationof buds.When the bud formed starch wasgradually consumed.The peroxidase activityincreased at formation of conidio-cell aggre-gate,then it reduced.When callus differen-tiate into bud,it increased again,when theshoot formed,it reduced again,so it formeda two peak curve.Isozyme analyses indicatedthat A_2 and A_3 are stable among four isoper-oxidase bands of the positive pole,A_1 andA_4 presented and changed regularly at thedifferent developmental stages.The contentsof soluble protein showed cyclic accumulation.The kind of acid proteins obviously increasedat the stage of dedifferentiation and redifferen-tiation.The relations between morphogeneticand changes of peroxidase and soluble proteinwere discussed.
