人胚肺成纤维细胞与人肺腺癌细胞共同培养时的形态学变化

摘    要:本文在混合培养的基础上建立了一种新的细胞培养技术。选用人肺腺癌细胞和人胚肺成纤维细胞,将两者按一定顺序定位培养在同一容器内的盖玻片上。在两种细胞相遇后,于不同时间取出长有细胞的盖片制成光镜和电镜标本。透射电镜标本采用了经过改进的原位包埋、水平切片法,使两种不同类型的细胞之间在定位培养时的超微结构变化得到充分显示。观察结果表明:两种细胞定位培养4天时,光镜观察未见明显的形态学改变,但电镜下已显示出变化;培养7天时,光镜及电镜观察均可见到与肿瘤细胞接触的成纤维细胞有损伤,与成纤维细胞接触的肿瘤细胞胞膜亦有破损。细胞化学染色显示肿瘤细胞周围的成纤维细胞内琥珀酸脱氢酶(SDH)活性降低。

Abstract:We have observed the morphological and histochemical changes of the pulmonary fibroblasts of humart fetus and human pulmonary adenocarcinoma cells cocultured in vitro withlight and electro microscope. A new cell culture method forlocation was employed to investigae the interaction between these two kinds of cells, which were cocultured in the same container designed by us. To prepare samples for TEM, embedding slice in situ and horizontalsectioning were adopted. It is convenient to observe the ultrastructure of different kinds ofcells wilh localized cell culture. The result suggested that the fibroblasts attached to cancercells changed unsignificantly on day 4 with light microscope. But the damage to the fibroblsts was obvious between day 4 and day 7 with electron microscope, and with histochemistrythe activities of their succinnate dehydrogenase (SDH) were seen decreased. The cytomembrane of the human pulmonary cancer cells were also damaged.
