D-半乳糖致衰老模型大鼠肝肾功能和自由基代谢与中药的干预作用

摘    要:目的观察D-半乳糖致衰老模型大鼠血糖血脂、肝肾功能和自由基代谢的变化,探讨中药的干预作用及其抗衰老的机制。方法大鼠每日皮下注射D-半乳糖,连续造模7周,建立大鼠衰老模型,造模同时,用抗衰老片和首乌延寿片进行干预,测定试验大鼠的血糖血脂、肝肾功能和SOD、MDA、Na+-K+-ATPase和Ca++-Mg++-ATPase等自由基代谢指标。结果造模7周后,模型对照组衰老大鼠TG、BUN明显升高(P〈0.05,P〈0.01),AST、ALP、CREA呈上升趋势(P〉0.05),肝、肾组织SOD活性明显降低(P〈0.05),肾组织MDA含量明显升高(P〈0.05),肝组织Na+-K+-ATPase和Ca++-Mg++-ATPase活性均显著降低(P〈0.05,P〈0.01);给予抗衰老片和首乌延寿片后,衰老大鼠TG、AST、BUN、CREA和UA均显著降低(P〈0.05,P〈0.01),肝、肾组织MDA含量显著降低(P〈0.05),SOD活性显著升高(P〈0.05,P〈0.01),肝组织Na+-K+-ATPase和Ca++-Mg++-ATPase活性均显著升高(P〈0.01)。结论D-半乳糖致衰老大鼠模型的血脂上升、肝肾功能异常和抗脂质过氧化的能力下降;给予抗衰老片和首乌延寿片后,可有效改善衰老大鼠的糖脂代谢和肝肾功能,提高肝肾组织的抗脂质过氧化的能力,提示中药的抗衰老作用可能与其抗氧化作用有关。

Abstract:Objective To observe the changes of glucose and lipid metabolism,the liver and renal function and the free radical metabolism of ageing rat model induced by D-galactose and to study the effect of traditional Chinese medicine on anti-ageing mechanism.Methods The ageing rat model was established by injecting D-galactose subcutaneously every day for seven weeks,meanwhile the rats in administration groups were given anti-ageing tablets and Shouwu Yanshou tablets respectively.At the end of the experiment,the glucose and lipid metabolism,the liver and renal function and SOD,MDA,Na +-K +-ATPase and Ca ++-Mg ++-ATPase were determined.Results After seven weeks,TG and BUN of the ageing rat in model control group significantly increased(P 0.05,P 0.01),the content of serum AST,ALP and CREA increased(P 0.05),SOD activity of the liver and renal tissue decreased significantly(P 0.05),the content of MDA of the renal tissue increased obviously(P 0.05),the Na +-K +-ATPase and Ca ++-Mg ++-ATPaseactivities of the liver tissue descended remarkably(P 0.05,P 0.01).After treatment of anti-ageing tablets and Shouwu Yanshou tablets,the content of serum TG,AST,BUN,CREA and UA significantly suppressed(P 0.05,P 0.01),the content of MDA of liver and renal tissue of ageing rats decreased remarkably(P 0.05),SOD activity increased remarkably(P 0.05,P 0.01),the Na +-K +-ATPase and Ca ++-Mg ++-ATPase activities of the liver tissue ascended remarkably(P 0.01).Conclusions The content of blood lipid of ageing rat model induced by D-galactose increases,the liver function and renal function are abnormal,the capability of anti-lipid peroxidation decreases,anti-ageing tablets and Shouwu Yanshou tablets can improve effectively the glucose and lipid metabolism and the liver and renal function,and enhance the capability of anti-lipid peroxidation of liver and renal tissue,which suggest that the anti-ageing effect of traditional Chinese medicine may be related to their antioxidation.
