单侧输尿管结扎再通大鼠模型的建立方法及其评价

摘    要:目的比较单侧输尿管结扎(UUO)与单侧输尿管结扎再通(RUUO)建立的SD大鼠肾纤维化模型的优劣,从而得到更符合临床慢性肾衰进展特点的动物模型,以供临床药物筛选、疗效评介、科研、教学使用。方法健康SPF级雄性SD大鼠54只,随机分成假手术组、UUO组及UUO再通组各18只,造模成功后检测血肌酐(Scr)、尿素氮(BUN)及尿N-乙酰-β-D-氨基葡萄糖苷酶(NAG),病理检查肾小管间质纤维化指数和免疫组化检查α-肌动蛋白(α-SMA)。结果无论从肾功能方面,还是病理检查方面,UUO再通的大鼠肾纤维化动物模型均优于UUO大鼠模型。结论再通UUO肾纤维化大鼠模型在功能及病理方面及其相关的免疫组化方面优于UUO组,其病理特点更符合慢性肾小管间质纤维化进程的动物模型。

Abstract:Objective To compare the model method between unilateral ureteral occlusion model(UUO) and recanalization of unilateral ureteral occlusion model(RUUO) so as to obtain a better fibrosis of kidney animal model to use in scientific research and teaching.Methods 54 SPF class SD male rats were randomly divided into three groups: normal group,UUO group,RUUO groups. In the end,contents of blood Scr and blood BUN urine n-acetyl-β-d-glucosaminidase(NAG) of every group were detected,degree of kidney fibrosis was assessed by the pathology of kidney tissue and α-smooth muscle actin immunohistochemistry of kidney tissue of every group.Results The contents of renal function and the pathology of kidney tissue and α-smooth muscle actin immunohistochemistry of kidney tissue results were obvious better in RUUO group than in UUO groups.Conclusion It is indicated that the model of RUUO established by recanalization is better than the UUO model.
