克山病病区粮喂养大鼠后心肌细胞自由钙浓度变化与硒的作用

摘    要:为了解克山病的发病机理,本文研究了低硒对心肌细胞钙转运的影响。用克山病病区粮喂养的大白鼠与用非病区粮喂养的大白鼠相比,心肌细胞胞浆自由钙的浓度高,心肌细胞膜流动性及Na~+,K~+-ATP酶活性也高,在病区粮中添加适量的硒,上述指标与用非病区粮喂养的大白鼠的差距缩小,说明低硒是引起心肌细胞钙转运失常从而使细胞浆自由钙浓度升高的重要因素但不是唯一的因素,文中讨论了细胞浆自由钙浓度与细胞膜流动性、Na~+,K~+-ATP酶活性及心肌线粒体功能的关系,以及低硒对它们的影响。

Abstract:In order to investigate the mechanism of Keshan Disease (KD) formation the present study of effects of Se-deficiency on the calcium transport in myocardial Cells was carried out.Results showed that in addition to the increase of cytosolic free calcium concentration in rats fed with KD area feed, the membrane fluidity and Na+,K+-ATPase activity of myocardial cells of the same rats were also increased compared with the rats fed with Beijing feed.It is worth to notice that by feeding the rats with the KD area feed supplemented with Na2SeO3 the changes in the three criteria mentioned above became less extensive compared with the rats fed with Beijing feed.It indicated that the deficiency in Se should be an important factor but not the only factor to cause the damage of calcium transport of myocardial cells thus resulting in the increase of cytosolic Ca+ concentration.It was discussed that relationship between the cytosolic Ca+ concentration and the membrane fluidity, Na+, K+ -ATPase activity and function of myocardial mitochondria, and the effects of Se-deficiency on them.
