鼻咽癌原代培养细胞分泌性蛋白图谱的初步研究

目的:建立细胞分泌性蛋白的蛋白质组学研究方法,在此基础上初步建立鼻咽癌细胞分泌蛋白图谱。方法:通过组织块法进行鼻咽癌细胞原代培养,收集细胞上清,超滤法脱盐并浓缩上清蛋白质,双向凝胶电泳技术建立细胞分泌性蛋白图谱。结果:鼻咽癌细胞原代培养取得成功,并初步建立了鼻咽癌细胞分泌性蛋白的双向凝胶电泳图谱。结论:建立了细胞分泌性蛋白的蛋白质组学研究方法,开展了鼻咽癌细胞分泌组学研究,为阐述鼻咽癌癌变机制提供了依据,也为后续研究打下了坚实的实验基础。

Preliminary Study of the Cell-secreted Proteomes of Nasopharyngeal Carcinoma

Objective:This study was designed to establish the two-dimensional electrophoresis profile of cell-secreted proteomics of nasopharyngeal carcinoma.Methods:Nasopharyngeal carcinoma were cultured by the technique of primary tissue culture in vitro,and cell-secreted protein from the cultured medium were harvested by ultrafiltrating concentration and desalina- tion.Samples were analyzed by two dimensional gel electrophoresis.Results:A suitable approach of primary culture in vitro of na- sopharyngeal carcinoma tissue was set up,and two dimensional gel electrophoresis diagram of cell-secreted protein was constructed. Conclusion:This research established a reliable method of cell secreted proteomics,and provided useful information and experi- mental basis for the study of nasopharyngeal carcinoma.
