摘    要:采用生物活性载体 ,通过吸附固定化方式 ,结合液体石蜡原位萃取技术 ,培养紫草细胞。测定了细胞生长、底物消耗和产物合成的动力学 ,紫草宁产率为 0 .916 g/g干重细胞和 0 .95 3g/g干重接种细胞 ,分别为悬浮培养的 12 .7倍和 6 .3倍。同时 ,对吸附与包埋固定化方法进行了综合比较 ,探讨了吸附固定化方法的应用前景。

Immobilization Culture of Lithospermum erythrorhizon Cells by Adsorption
Abstract:Lithospermum erythrorhizon was immobilized by adsorption using bio-active carrier made from fungal cell mycelia through a series of physiochemical treatment. Then immobilized cell culture was conducted by in situ extraction with liquid paraffin. In this paper, the dynamic research focused on cell growth, substrate consumption and product secretion. It was found that cell growth was greatly reduced while the shikonin productivity significantly increased by immobilization culture by adsorption. The shikonin productivity in adsorption culture was 0.916 g/g dry cell weight or 0.953 g/g dry cell weight inoculation, which was 12.7 times and 6.3 times higher than that in suspension culture, respectively(Table 2). Also, the effects of the use of two immobilization methods, adsorption and entrapment, on cell growth and shikonin productivity were compared. The potential application of immobilization by adsorption on large-scale cell culture was further discussed.
