玉米种胚萌发过程中钙调素含量变化及与赤霉素和脱落酸的关系

摘    要:用对植物钙调素具有专一性的小麦钙调素抗体建立了定量植物钙调素的酶联免疫吸附测定法。该方法的线性测定范围为2至1400ng;批内与批间变异系数分别为7.9和10.6。用此方法对玉米种胚的测定结果表明,钙调素含量随着萌发时间延长而增加。赤霉素在促进玉米种子萌发的同时,种胚中的钙调素含量也提高;脱落酸抑制玉米种子萌发,同时种胚中的钙调素含量也降低。这些结果说明钙调素可能在种子萌发过程中具有重要作用。

Abstract:Using antibody against native wheat calmodulin,a competitive indirect enzvme-linkedimmunosorbent assay(ELISA)was developed for the quantitation of calmodulin in plants.The linear range of calibration ctirve was between 2 and 1400 ng calmodulin.The meanCV% of ELISA in intra-assay and inter-assay were 7.9% and 10.6% respectively.The levelof calmodulin in germinating corn seed embryos were measured by ELISA.During the ger-mingation process,calmodulin content of per fresh weight and per mg protein of embryos in-creased.The increase of calmodulin is promoted correspondingly with germination promotionby gibberellic acid(GA_3);and the increase of calmodulin is blocked by abscisic acid(ABA),which simultaneously also blocks germination.These results suggest that calmodulin couldplay an important role in the regulation of germination of corn seeds.
