摘    要:酰基辅酶A硫酯酶(acyl-CoA thioeaterase,ACOT)属于广泛的硫酯水解酶的一类酶,分属于α/β水解酶超家族和"HotDog"超家族。ACOT作用范围很广,可以裂解支链脂肪酸、短链和长链饱和及不饱和酰基辅酶A等多种含辅酶A的底物,在脂肪酸代谢途径中发挥信号转导、基因调控等生理生化作用。因此研究ACOT的结构和功能,可以为某些疾病治疗和发病机理研究提供依据,也可以为指导现有的代谢途径和重新设计新的合成途径来合成天然或非天然产物提供依据和基础。

Abstract:Acyl-CoA thioeaterase(ACOT),a wide range of thioester hydrolases,belongs toα/βhydrolase superfamily and“HotDog”superfamily,respectively.ACOT has a wide range of functions,which can cleave branched-chain fatty acids and various substrates containing coenzyme A such as short-and long-chain saturated and unsaturated acyl-CoA(CoASH).In the fatty acid metabolism pathway,ACOT plays physiological and biochemical functions in signal transduction and gene regulation.Therefore,the studies on the structure and function of ACOT can provide a basis for diseases therapy and pathogenesis research,and can also guide the existing metabolic pathways and redesign new synthetic pathways to synthesize natural or non-natural products.
