从短锯鲉分离和鉴定抗冻多肽mRNA

摘    要:短锯鲉在冬季产生抗冻多肽,此多肽能降低血液的冻结温度。本实验用11月份捕获的短锯鲉肝脏做标本,提取抗冻多肽mRNA,经寡聚脱氧胸腺嘧啶核苷酸纤维素亲合层析和蔗糖梯度密度离心提纯。抗冻多肽mRNA的长度经含羟甲基汞的琼脂糖凝胶电泳测定为580硷基对,其在无细胞翻译系统中初级翻译产物的分子量经SDS-PAGE估计为15000。

Abstract:The messenger RNA coding for the serum antifreeze polypeptide of the Shorthorn Sculpin,Myoxocephalui Scorpins,was extracted from the liver polysomes of fish caught in November and was purified by oligo(dT)-cellulose chromatography and sucrose gradient centrifugation.The length of the mRNA was 580 nucleotides measured by agarose gel electrophoresis in the presence of methyl mercury.In the cell-free tianslation system the molecular weight of its primary translation product was 15000 estimated by sodium dodecyl sulfate gel electrophoresis.
