壳寡糖对肝癌细胞SMMC-7721中Bcl-2和Caspase-3表达的影响

摘    要:目的检测壳寡糖对人肝癌SMMC-7721细胞的抑制效果及对凋亡调控蛋白Bcl-2和Caspase-3的影响。方法采用噻唑蓝(MTT)法检测不同浓度壳寡糖对肝癌细胞SMMC-7721细胞增殖的抑制作用,并利用荧光Hoechst33258染色法检测细胞凋亡状况。最后通过免疫细胞化学方法研究壳寡糖对肝癌细胞SMMC-7721中Bcl-2和Caspase-3表达的影响。结果壳寡糖能够抑制SMMC-7721细胞增殖,并且促进SMMC-7721细胞的凋亡,并且壳寡糖能够上调促凋亡蛋白Caspase-3的表达和降低抑制凋亡蛋白Bcl-2的表达。结论壳寡糖对人肝癌SMMC-7721细胞的增殖有抑制作用,此作用可能是通过促进Caspase-3的表达和抑制Bcl-2的表达来实现的。

Abstract:Objective To investigate the inhibitory function of oligochitosan on human hepatocelular carcinoma cell line SMMC-7721 and determine the effect of oligochitosan on the expression of Caspase-3 and Bcl-2 in the hepatoma cell line. Method MIT assay was applied for evaluation of proliferation of SMMC-7721 treated by different concentrations of oligochitosan. Hoechst 33258 coloration was adopted to detect cell apoptosis. Immunocytochemistry was utilized to measure the expression of Caspase-3 and Bcl-2 in the SMMC-7721 cells treated by oligochitosan. Result Oligochitosan inhibited the proliferation of SMMC-7721 cells and accelerated cell apoptosis. The expression of Caspase-3 and Bcl-2 were respectively higher and lower in oligochitosan treated group than those in the control group. Conclusion The mechanisms of oligochitosan inhibiting the proliferation of SMMC-7721 cells were possibly that oligochitosan up-regulates the expression of Caspase-3 and down-regulates the expression of Bcl-2.
