裙带菜多糖及其寡糖对 洛哌丁胺诱导便秘小鼠的肠道菌群的影响

摘    要:目的比较裙带菜多糖及其寡糖对洛哌丁胺引起小鼠便秘的肠道菌群的影响。方法给予小鼠9.6mg/kg浓度的洛哌丁胺每日2次连续灌胃14d,分别给予裙带菜多糖和裙带菜寡糖,通过PCRDGGE技术获得菌群指纹图谱和16SRNA测序,用Quantity One软件构建进化树,对图谱进行相似性及多样性分析,并选取差异条带进行测序。结果裙带菜多糖可以明显改善小鼠由洛哌丁胺引起的便秘并且增加菌群多样性,裙带菜寡糖虽然也可以改善但效果不是很明显(对便秘和菌群有改善,但差异无统计学意义)。结论裙带菜多糖及其寡糖可以(通过改善肠道菌群从而)改善由洛哌丁胺引起的便秘。

Effects of wakame polysaccharides and its oligosaccharides on intestinal flora of mice with constipation induced by Loperamide
Abstract:Abstracts: Objective To compare the effects of wakame polysaccharides vs its oligosaccharides on intestinal flora of mice with Loperamide-induced constipation. Methods The mice were given 9.6 mg/kg of Loperamide twice daily. After 14 days of continuous gavage, wakame polysaccharides and wakame oligosaccharides were given respectively. The fingerprint of the flora and 16S RNA sequence were obtained by using PCR-DGGE technique. The phylogenetic tree was constructed using Quantity One software, and the similarity and diversity of the flora were analyzed. The differential bands were selected for sequencing. Results Wakame polysaccharides significantly relieved the constipation caused by Loperamide and increased the diversity of the flora. Wakame oligosaccharides also showed an relieving effect but not as obviously as wakame polysaccharides did (the constipation was relieved and the flora were improved without significant differences). Conclusion Wakame polysaccharides and its oligosaccharides can relieve constipation caused by Loperamide by improving intestinal flora.
