植物生物质生物降解组合菌株的筛选研究

摘    要:本研究用平板混合培养和实际降解相结合的方法筛选了较优的降解植物生物质的菌株组合。首先将6株纤维素降解菌和7株木质素降解菌两两平行划线接种到基础平板上,通过观察菌落形态,挑选出了9对能较好共存的菌株组合。随后对这9个组合进行实际降解稻草的研究,培养结束后体系中半纤维素、纤维素和木质素的含量分析结果表明AF93252 M5和XP M5这两个菌株组合较理想,它们混合培养时对稻草的降解效果均优于其单独降解稻草的效果。

Abstract:Some combined strains for degradation of straw were screened from some cellulolytic and ligninolytic strains stored in our laboratory by means of plate mixed cultivation and actual degradation.Firstly,combinations of thirteen strains prepared for study were pair wise inoculated into the nutrient agar plates by parallel streaking.By observing the colonial morphology of these strains,nine couples of strains were found that they could grow well with each other.Then,these couples were examined by solid-state fermentation with shredded straw.After fermentation,the hemicelluloses,cellulose and lignin contents of residues were measured respectively.The results indicated that the combined strains AF93252 M5 and XP M5 were the ideal couple because it is of higher efficiencies in mixed culture than that in pure culture.This simple and highly efficient method on screening combined strains was proved to be useful for studying how to improve the efficiency of straw biodegradation.
