摘    要:目的:探讨核转录因子NFIL6对肝癌细胞系BEL7404恶性度的影响。方法:用磷酸钙介导转染技术,将NFIL6表达载体(pCN)和空载体质粒(pCN空)分别导入肝癌细胞系BEL7404,并借助细胞生长曲线,软琼脂集落形成试验,裸鼠成瘤试验对转染细胞的恶性度进行了检测。结果:与原细胞系BEL7404和空载体转染的该细胞系相比较,转染了NFIL6基因的BEL7404的各细胞克隆生长速度减慢,在软琼脂中集落形成率恶性度下降,裸鼠成瘤试验显示成瘤性明显降低。结论:表明外源转染的NFIL6对肝癌细胞系BEL7404具有明显的肿瘤抑制作用 。

Abstract:NF IL6 (Nuclear factor for IL 6 expression) is involved in inflammatory reactions,expression of acute phase proteins and cytokines,apoptosis and suppression of tumors.To investigate the role of expression of exogenous NF IL6 in tumors,a recombinant expression plasmid,pCN,which harbored the coding region of NF IL6 ,was transfected into the human BEL 7404 hepatoma cell line by calcium phosphate transfection,and the transfectants stably expressing NF IL6 gene were isolated by G418 screening.Then cell proliferation assay,soft agar assay and nude mice tumorigenesis assay were performed to examine the effect of NF IL6 expression on cell growth and tumor formation.RESULTS:BEL 7404 transfectants stably expressing full length coding region of NF IL6 were established by calcium phosphate mediated transfection and selected for further study.When cell proliferation was followed over several days,it was found that the NF IL6 transfectants generally grew more slowly than nontransfected cells.And the NF IL6 transfectants exhibited markedly lower colony formation rate.The NF IL6 transfectant clones were injected into Balb/c nu/nu nude mice,the sizes of outgrown tumors were measured in every third days,and after 40 days the animals were sacrificed and the tumors were dissected.It was found that the tumor volumes formed in mice from NF IL6 transfectant clones were generally significantly smaller than the tumors formed from control cell lines.CONCLUSION:These facts indicate that the expression of exogenous NF IL6 in human BEL 7404 hupatoma cell line has the potential to suppress growth and tumorigenesis of these cancer cells.
