Abstract:A method of amino acid analysis of proteins and peptides through hydrolysis in a microcapillary tube with methanesulfonic acid containing dithiediglycolic.acid is reported. Hydrolysis in a microcapillary tube eliminates the necessity of time-consuming sealing under vacuum and flushing with nitrogen- High yield of derivatization of cysteine or half-cystine by dithiodiglycolic acid has been achieved and the recovery of tryptophan is greater than 90% by this method.
